abstract

background: free fetal dna (ffd) in maternal plasma/serum has increasingly become the source of fetal material for diagnostic purposes in recent years. this source of fetal material can be used for sex determination. rh typing paternally inherited sequences and compound heterozygosity. reports on the lack of consistent pcr amplification of y-chromosome sequences of ffd in maternal plasma/serum have led diagnostic services to the use of real-time pcr for improved sensitivity of sex determination. we report the use of conventional pcr for fetal sex determination with high sensitivity and reporoducibility.  methods: peripheral blood samples were obtained from 21 pregnant volunteers during 4-29 weeks of gestation. a healthy man and 52 healthy non-pregnant women were investigated in this study as controls. all the samples were collected at random. fetal gender was determined by conventional pcr to detect a y-chromosomal sequence (dyz1) in maternal serum and confirmed by amniocentesis or after delivery.  results: fetus-derived y sequences were detected in 16 out of 17 maternal serum samples with male fetuses and none of the 4 women bearing female fetuses had positive results. the sensitivity of our pcr reached >95%, and the specificity reached >96% (y signal detected in only 2 out of 52 female samples studied).  conclusion: our report on the improved sensitivity of y- chromosome pcr simplifies routine sex determination in diagnostic laboratories without the use of real-time pcr.

early non-invasive determination of fetal sex using cell-free dna

the first step in the prenatal diagnosis of x-linked genetic disorders is determining fetus gender. current invasive methods to obtain the dna source of the fetus instead of its miscarriage risk, has harmful stress for high risk pregnancies. cell free fetal dna (cffdna) circulating in the maternal blood, has now become a useful source of noninvasive prenatal diagnosis. considering limitation of...

fetal sex determination using non-invasive method of cell-free fetal dna in maternal plasma of pregnant women during 6th– 10th weeks of gestation

in previous years, identification of fetal cells in maternal blood circulation has caused a new revolution in non-invasive method of prenatal diagnosis. low number of fetal cells in maternal blood and long-term survival after pregnancy limited the use of fetal cells in diagnostic and clinical applications. with the discovery of cell-free fetal dna (cffdna) in plasma of pregnant women, access to...

Prenatal determination of farm animal fetal sex using free fetal DNA in maternal plasma

In many farm animal species, the preference of a specific sex is obvious mainly due to commercial implications; so for comprehensive and efficient results in the livestock management plans specially breedingand selection programs, it is necessary to include acost-effective and reliable strategy for early determination of fetal sex. Several invasive and non-invasive approaches have been establis...

A multiplex PCR for non-invasive fetal RHD genotyping using cell-free fetal DNA.

AIM To design a protocol for non-invasive prenatal diagnosis of fetal Rhesus D (RhD) status. MATERIALS AND METHODS A total of 112 single lymphocytes were used to test the efficiency of the assay. The protocol was validated using blood samples from 84 RhD-negative pregnant women at 7-24 weeks of gestation. Cell-free DNA (cfDNA) was enzymatically digested using AciI and analyzed by a polymerase...

Non-invasive prenatal diagnosis of fetal trisomy 21 using cell-free fetal DNA in maternal blood

Since the existence of cell-free fetal DNA (cff-DNA) in maternal circulation was discovered, it has been identified as a promising source of fetal genetic material in the development of reliable methods for non-invasive prenatal diagnosis (NIPD) of fetal trisomy 21 (T21). Currently, a prenatal diagnosis of fetal T21 is achieved through invasive techniques, such as chorionic villus sampling or a...

Non-invasive prenatal testing using cell-free fetal DNA in maternal circulation.

The identification of cell-free fetal DNA (cffDNA) in maternal circulation has made non-invasive prenatal testing (NIPT) possible. Maternal plasma cell free DNA is a mixture of maternal and fetal DNA, of which, fetal DNA represents a minor population in maternal plasma. Therefore, methods with high sensitivity and precision are required to detect and differentiate fetal DNA from the large backg...
